چکیده
AIM To compare magnetic resonance imaging (MRI), micro-computed tomography (μCT), optical reflectance imaging (ORI) and caliper measurements for subcutaneous tumor detection and size assessment. MATERIALS AND METHODS HCT 116-green- (GFP)-red-fluorescent protein (RFP) tumor volumes were measured in vivo by calipers and by ORI, MRI and μCT over 15 days and validated ex vivo. The method correlating best with the ex vivo tumor volumes was used as reference for longitudinal in vivo correlations. RESULTS MRI and ORI detected tumors at day 1 post-injection, μCT after 3 days. The in vivo MRI data correlated best with the ex vivo tumor volumes (r²=0.96), followed by μCT (r²=0.93). Thus, MRI was chosen as the reference. μCT-(r²=0.90), in vivo caliper data (r²=0.80) and fluorescence intensities (GFP:r²=0.71; RFP:r²=0.75) highly correlated with MRI-data, whereas fluorescent areas (GFP:r²=0.26; RFP:r²=0.30) poorly correlated. CONCLUSION MRI sensitively detects tumors and precisely determines their size; μCT is an accurate alternative for larger tumors; ORI is as sensitive as MRI, but overestimates small tumor sizes; and fluorescence intensity correlates better with tumor volume than fluorescence area.

Color-coded fluorescent protein imaging of angiogenesis: the AngioMouse models.
We have utilized multicolored fluorescent proteins to develop three imaging models of tumor angiogenesis. In one model, the nonluminous induced capillaries are clearly visible by contrast against the very bright tumor green fluorescent protein (GFP) fluorescence examined either intravitally or by whole-body imaging in real time. Intravital images of an orthotopic model of human pancreatic tumor...

The use of green fluorescent protein (GFP) for detection and visualization of cells in vivo has been established by our laboratory. With this fluorescent tool, tumor cells were detected and visualized for the first time at the microscopic level in fresh viable tissue in their normal host organ (1). The use of GFP in whole-body imaging was also established by our laboratory (2). Fluorescent tumo...
متن کاملDual-color fluorescence imaging distinguishes tumor cells from induced host angiogenic vessels and stromal cells.
We have developed a simple yet powerful technique for delineating the morphological events of tumor-induced angiogenesis and other tumor-induced host processes with dual-color fluorescence. The method clearly images implanted tumors and adjacent stroma, distinguishing unambiguously the host and tumor-specific components of the malignancy. The dual-color fluorescence imaging is effected by using...
متن کاملClinically-relevent orthotopic metastatic models of pancreatic cancer imageable with fluorescent genetic reporters.
This article describes authors' cumulative experience with the development and preclinical application of clinically-relevant, metastatic orthotopic mouse models of pancreatic cancer made imageable with genetic reporters. These models utilize the human pancreatic cancer cell lines which have been genetically engineered to selectively express high levels of green fluorescent protein (GFP) or red...
متن کاملDynamic Contrast Magnetic Resonance Imaging (DCE-MRI) and Diffusion Weighted MR Imaging (DWI) for Differentiation between Benign and Malignant Salivary Gland Tumors
Background: Salivary gland tumors form nearly 3% of head and neck tumors. Due to their large histological variety and vicinity to facial nerves, pre-operative diagnosis and differentiation of benign and malignant parotid tumors are a major challenge for radiologists. Objective: The majority of these tumors are benign; however, sometimes they tend to transform into a malignant form. Functional M...
